We've seen other delivery ponies so it wouldn't be too much of a stretch to assume there is more than one pony delivering mail, but she could be.

I have not seen other delivery ponies So I'm guessing that Spike's way of mailing things is like e-mail today: some people use it, but for those who don't, they get pegasi mail.

I would imagine that conjured muffins are not Derpy's sole food source...
There are no other dragons that look like Spike, we don't see any other dragons at all in Canterlot, I think his magicking is an (expensive) royal privilege. There are other delivery ponies in that Feeling Pinkie Keen segment, although it's unclear what their structure is. Although someone else raised a point that I never thought about: think about that scene in CMC logically: what the hell is the deal with the egg? Do they raid dragon nests? How many tests are they giving, is it supposed to be like the Kobayashi Maru? I would guess dragons have at least a slight magic resistance, but do mothers really willingly donate their unborn children to be subjected to the blasts of young unicorns?
